Effects of Methylsulfonylmethane and Epidermal Growth Factor Receptor-Tyrosine Kinase Inhibitor on Cell Proliferation, Cell Cycle and Anti-Inflammatory Effect in Chicken Intestinal Epithelial Cells with Lipopolysaccharide Challenge

Background: Intestinal epithelial injury stress is one of the most common stresses in contemporary poultry breeding. This study aims to evaluate protective effects Methylsulfonylmethane (MSM) and epidermal growth factor receptor-tyrosine kinase inhibitor (EGFR-TKI, erlotinib) on induced by lipopolysaccharides (LPS) chicken intestinal cells (IECs). Methods: Cell Counting Kit-8 (CCK-8) EdU assays were used cell viability proliferation. The cycle was analyzed flow cytometry method. mRNA transcript protein expression levels EGFR detected RT-qPCR Western blotting (WB). pro-inflammatory cytokines determined using ELISA kits. Result: results have shown that MSM could promote proliferation regulate arrest when inhibited (P less than 0.05). In addition, both EGFR-TKI decrease expressions IL-1β, IL-6 TNFα These showed beneficial roles IECs LPS may be related promoting inhibiting inflammation; meanwhile, our experimental provide valuable information for further possible mechanism improve function regulating signaling pathway.
